Plasma levels of miR-19-5p correlate with sFLT-1 levels in gestational diabetes mellitus

چکیده
Background: Gestational Diabetes Mellitus (GDM) is a major complication of pregnancy that can seriously affect the health of mother and fetus. sFlt1 and miR-19-5p have been proved to be involved in the complications of GDM. Our study aimed to investigate the effects of sFlt1 and miR-19-5p on the complications of GDM. Method: Pregnant women with GDM who gave birth to normal babies and babies with macrosomia, and Fetal Growth Restriction (FGR) as well as normal pregnant women were included. Levels of soluble fms-like tyrosine kinase-1 (sFlt-1) and miR-195-5p in prepartum serum and postpartum placental samples were detected, and level of sFlt1 in Human Umbilical Vein Endothelial Cells (HUVECs) transfected with miR-195-5p was also measured. Results: GDM pregnant women that delivered normal and FGR babies had higher levels of both miR-195-5p and sFlt1 compared with GDM pregnant women who delivered macrosomia babies or normal pregnant women. Transfection with miR-195-5p decreased expression level of sFlt1 and decreased level of p-VEGFR2 in HUVECs, leading to inhibition of VEGFR2-mediated neonatal angiogenesis. It was also confirmed that the placental blood vessel density was decreased, leading to

Central nervous system development-related microRNAs levels increase in the serum of gestational diabetic women during the first trimester of pregnancy
MicroRNAs are heterochronic molecules important during brain development, which could be altered by gestational diabetes mellitus (GDM). To explore these molecules in maternal serum, we performed an RT-qPCR analysis. Our results revealed the heterochronic character of some neural development-related microRNA in serum samples of pregnant women. In relation to the first trimester, higher levels o...
متن کاملMiR-21-5p and miR-126a-3p levels in plasma and circulating angiogenic cells: relationship with type 2 diabetes complications
Innovative biomarkers are required to manage type 2 diabetic patients (T2DM). We focused our study on miR-126-3p and miR-21-5p levels, as biomarkers of endothelial function and inflammation. MiRNAs levels were measured in plasma from 107 healthy subjects (CTR) and 193 diabetic patients (T2DM), 76 without (T2DM NC) and 117 with (T2DM C) complications. When diabetic complication were analysed as ...
متن کاملPeriodic assessment of plasma sFlt-1 and PlGF concentrations and its association with placental morphometry in gestational hypertension (GH) - a prospective follow-up study
BACKGROUND Hypertensive disorders in pregnancy contributes to about 12% of maternal deaths in Malaysia and similarly worldwide. Early detection and adequate management are preventable strategies. Biochemical markers of abnormal angiogenesis would be more specific in early detection than routine blood pressure and proteinuria measurements. The aim of this study was to estimate maternal plasma Pl...
متن کاملMaternal serum lipid levels in pregnant women with gestational diabetes mellitus (GDM) in comparison to normal pregnant women
Background: Even though hormonal changes in pregnancy have been associated with plasma lipid variation, is not yet understood the mechanism which pregnancy alters lipid metabolism. Materials and Methods: This prospective cohort study conducted between March 2011 and May 2012 in Shariati Hospital on 112 pregnant women with GDM and 159 healthy pregnant women. In order to determine lipids or li...
متن کاملA MicroRNA Signature in Gestational Diabetes Mellitus Associated with Risk of Macrosomia.
BACKGROUND/AIMS MicroRNA (miRNA) is a small non-coding RNA molecule that functions in regulation of gene expression by targeting mRNA to affect its stability and/or translation. The aim of this study was to evaluate the miRNAs involvement in gestational diabetes mellitus (GDM), a well known risk factor for fetal overgrowth. METHODS Differential microRNA expression in placental tissues of norm...
